まず私は、細胞質再配置メカニズムへの関与が期待されるホヤ母性タンパク質p58の分子的実体がF0F1-ATPaseのαサブユニット(ATPα)と同一あることを明らかにした。一般的に、F0F1-ATPaseはミトコンドリア内膜に局在してATPを産生するタンパク質複合体として知られている。しかし、ホヤ卵ではF0F1-ATPaseのαおよびβサブユニットだけが、ミトコンドリア外のマイオプラズム表層領域に局在し、ミトコンドリア内とは異なる構成の複合体を形成して細胞骨格と結合していた。この新規な複合体をexo-ATPα/βと名付けた。\n さらに、ミトコンドリア呼吸鎖阻害剤として知られるsodium azide (NaN3)が、微小管依存的な第二細胞質再配置を阻害することを明らかにした。別の呼吸鎖阻害剤であるoligomycinには同様の阻害効果が見受けられず、NaN3が呼吸鎖以外の分子を阻害したことがわかった。また、NaN3が卵割期胚におけるspindle midzoneの形成やspindle orientationといったspindle dynamicsに関わる微小管依存的メカニズムも阻害することがわかった。Spindle dynamicsに対する同様な阻害効果は、ウニ胚やヒト培養細胞においても見受けられ、NaN3が異なる動物種間で保存されている分子をターゲットにして、微小管依存的な共通メカニズムを阻害していることが示唆された。\n 本研究で明らかにしたexo-ATPα/βの存在は、常識的なミトコンドリア内膜のF0F1-ATPaseに対する理解を根底から覆し、細胞生物学をはじめとした様々な分野に多大なインパクトを与えるものである。さらに、今後のexo-ATPα/βの構成分子および機能解析とNaN3のターゲット分子の同定は、細胞内物質輸送の新たなメカニズムを明らかにするだけでなく、細胞分裂におけるspindle
